Breast cancer ranks first as the highest incidence of cancer surpassing lung cancer in Indonesia. HER2+ as one of the subtype of breast cancer accounts for poor prognosis in patients as well as developing resistance to first line treatment known such as trastuzumab. Aaptamine is secondary metabolite compound derived from Aaptos suberitoides has been discovered to have anti-cancer activity. This study aimed to investigate the cancer cell growth inhibition effect on 3D breast cancer cell culture (spheroid assay). HCC1954 breast cancer cell line were used as spheroids representing trastuzumab-resistant HER2+ breast cancer cell. Spheroids were grouped into negative control and serial treatment group consisting 40 &micro;M, 80 &micro;M, and 120 &micro;M concentration of aaptamine. Cancer cell growth inhibition was analyzed using spheroid growth assay observed under inverted microscope with 100x magnification on day 0, 4th and 8th. This study revealed that there was statistically significant size reduction of spheroids treated with 80 &micro;M and 120 &micro;M compared to control group on 8th day (p value < 0.05).
