dc.description.abstractABSTRAK Latar Belakang: Karsinoma sel skuamosa rongga mulut pada dasarnya merupakan penyimpangan gen yang menimbulkan proliferasi berlebihan dan progresif, terjadinya penurunan apoptosis dan bertahannya hidup sel dikarenakan adanya gangguan pada pengaturan dan koordinasi pertumbuhan dan diferensiasi sel, termasuk peningkatan proliferasi akibat peningkatan ekspresi Cyclin D1 dan penghambatan apoptosis akibat protein antiapoptosis Bcl-2. Kurkumin (Curcuma domesticae) diketahui memiliki efek antikanker, salah satunya melalui mekanisme antiproliferasi dan pro apoptosis. Pemberian kurkumin akan mencegah terjadinya proses karsinogenesis dengan menurunkan ekspresi Cyclin D1 dan Bcl-2 Tujuan Penelitian: Penelitian dilakukan untuk menganalisis efek terapi kurkumin pada tahap dysplasia epitel di model karsinoma sel skuamosa rongga mulut melalui imunoekspresi Cyclin D1 dan Bcl-2. Metode: Penelitian merupakan penelitian eksperimental murni yang dilakukan pada binatang percobaan tikus Sprague dawley untuk menganalisis pengaruh pemberian kurkumin pada model karsinoma sel skuamosa rongga mulut tahap displasia epitel yang diinduksi dimethylbenz(a)anthracene (DMBA) 0,5% selama 4 minggu dengan pemeriksaan imunoekspresi Cyclin D1 dan Bcl-2. Hasil Penelitian: Terdapat perbedaan imunoekspresi Cyclin D1 dan Bcl-2 pada model karsinoma sel skuamosa rongga mulut tahap displasia epitel antara kelompok yang diberi kurkumin dan kelompok yang tidak diberi kurkumin per oral. Kata Kunci: displasia epitel pada karsinoma sel skuamosa rongga mulut,, kurkumin, DMBA, Cyclin D1, Bcl-2 ABSTRACT Background: Oral Squamous cell carcinoma (OSCC) is essentially an aberration of genes that cause excessive and progressive proliferation, decrease apoptosis and increase cell survival due to interference with coordination of the growth and cell differentiation, including increasing proliferation due to increased expression of Cyclin D1 and inhibition of apoptosis due to antiapoptosis Bcl-2. Curcumin (Curcuma domesticae) are known to have anticancer effects, one through the mechanism of anti-proliferation and pro-apoptosis. Curcumin can prevent carcinogenesis process by reducing the expression of Cyclin D1 and Bcl-2. Purposes: The study is conducted to analyze the effect of curcumin therapy in OSCC epithelial dysplasia stage through immunoexpression of Cylin D1 and Bcl-2. Method: This study is a true experimental study to analyze the effect of curcumin on the model of OSCC epithelial dysplasia stage induced by dimethylbenz (a) anthracene by analyzing immunoexpression level of Cyclin D1 and Bcl-2. Result: There are differences in imunoexspression level of Cyclin D1 and Bcl-2 on the model of OSCC epithelial dysplasia stage induced by dimethylbenz (a) anthracene (DMBA) 0,5% between the group given curcumin orally with the group not given curcumin.
