UJI AKTIVITAS ANTIBAKTERI MINYAK ATSIRI DAUN JINTEN (Coleus amboinicus Lour.) TERHADAP BAKTERI Methicillin Resistant Staphylococcus aureus (MRSA) DAN Pseudomonas aeruginosa Multi Resistant (PAMR) DENG

Abstract

Salah satu tumbuhan di Indonesia yang memiliki kandungan minyak atsiri sebagai antibakteri adalah daun jinten (Coleus amboinicus Lour.). Penelitian sebelumnya menunjukkan bahwa minyak atsiri daun jinten mempunyai aktivitas antibakteri yang kuat terhadap Staphylococcus aureus dan Pseudomonas aeruginosa tetapi belum pernah dilaporkan untuk bakteri Staphylococcus aureus dan Pseudomonas aeruginosa yang resisten. Penelitian ini dilakukan untuk mengetahui aktivitas antibakteri minyak atsiri daun jinten (Coleus amboinicus Lour.) terhadap bakteri Methicillin Resistant Staphylococcus aureus (MRSA) dan bakteri Pseudomonas aeruginosa Multi Resistant (PAMR), menentukan konsentrasi hambat tumbuh minimum (KHTM), dan menentukan konsentrasi bunuh minimum (KBM). Pengujian aktivitas antibakteri, penentuan KHTM dan KBM dilakukan dengan metode mikrodilusi. Hasil pengujian aktivitas antibakteri minyak atsiri daun jinten menunjukkan bahwa minyak atsiri daun jinten memiliki aktivitas antibakteri terhadap bakteri resisten tersebut. KHTM minyak atsiri daun jinten terhadap MRSA terletak pada konsentrasi 750 µg/mL dan KBM terletak pada konsentrasi 1000 µg/mL. KHTM minyak atsiri daun jinten terhadap bakteri PAMR terletak pada konsentrasi 375 µg/mL, dan KBM terletak pada konsentrasi 750 µg/mL. Hasil Gas chromatography–mass spectrometry (GCMS) menunjukkan bahwa minyak atsiri daun jinten mengandung Carvacrol, Caryophyllene, Bergamotene, dan α Humulene. One of the plants in Indonesia that contains oil as an antibacterial is Jinten leaves (Coleus amboinicus Lour.). Previous research showed that essential oil of Jinten leaves has a strong antibacterial activity against Staphylococcus aureus and Pseudomonas aeruginosa but has not been reported for the bacteria Staphylococcus aureus resistant and Pseudomonas aeruginosa resistant. This research was already conducted to determine the antibacterial activity of Jinten leaves essential oil against the Methicillin Resistant Staphylococcus aureus (MRSA) bacteria and Pseudomonas aeruginosa Multi Resistant (PAMR) bacteria, determine the Minimum Inhibitory Concentration (MIC), and determine the Minimum Bactericidal Concentration (MBC). Test for antibacterial activity, MIC and MBC determined by microdilution method. The results revealed that the essential oil of Jinten leaves have antibacterial activity against these resistant bacteria. The MIC of the Jinten leaves essential oil to MRSA was in concentration of 750 µg/mL and MBC was in concentration of 1000 µg/mL. MIC of the Jinten leaves essential oil to PAMR bacteria was in concentration 375 µg/mL, and MBC was in concentration of 750 µg/mL.
